On the first play-off hole, both players made par in different ways, but Henseleit's drive landed in a gorse bush on the second hole, leaving her unable to save par and handing Kuwaki the victory.
Kuwaki's Historic Win and Journey
Kuwaki, the 43rd different winner in 50 editions of the tournament, said: 'It's an amazing feeling to win again as a Japanese player. I hope as team Japan we can continue to light up the golf world.' Her previous five professional wins had all come on her home tour.
The 27-year-old won just 12 months after failing to make the cut at Royal Porthcawl, marking a remarkable turnaround. She will represent Europe at the Solheim Cup in September, adding to her growing international profile.
Noh's Heartbreak and Other Contenders
Yealimi Noh finished one stroke behind Kuwaki, becoming the first player to hold a three-stroke lead after 54 holes and fail to win the Women's Open. Her hopes of a second professional victory were dashed on the blustery Lancashire coast.
World number one Nelly Korda ended at two under after a double-bogey on Saturday's third round, while Lottie Woad and Charley Hull's English challenge failed to spark. Haeran Ryu's contention was hampered by frequent bunker trips.
